| Abstract | DNA microarray technique is currently one of the most widely used methods for genome scale expression analysis with numerous applications. For DNA microarray analysis of the gene expression of small number of cells, amplification of RNA is often required. In this study, we evaluated the accuracy and reliability of the RNA amplification method typically used for DNA microarray experiment. Using the limited quantities of Peripheral Blood Mononuclear Cells (PBMCs), RNA was extracted and amplified. Then we performed DNA microarray experiment using highdensity DNA microarray platform. We describe here the critical factors and parameters that we observed and expect that the information provided here will be a useful guide for researchers designing similar experiments. |
