Title Development of fibrinogen microspheres as a biodegradable carrier for tissue engineering

Abstract Protein based biomaterials from plant or animal sources are widely used in drug delivery, wound dressing, and tissue engineering applications, due to their enhanced biocompatibility and specific functions. On the other hand, their lower mechanical strength and fast rate of degradation were often required reinforcements with the protein carriers or biodegradable polymers. To overcome above limitations, the cross-linked fibrinogen (Fbg) microspheres were developed using water soluble1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC). The Fbg microspheres (Fbg-MP) were produced from various EDC concentrations (1, 3, 5 and 10 mM). The physical characterizations of Fbg-MP were carried out with SEM, FTIR, TGA, DSC and XRD measurements. In addition, swelling test and fibrinolysis assay with proteases indicated that cross-linked Fbg-MPs were designed to be resistant to the solvents and proteases, where their strength were strongly depended on the concentrations of EDC. The biocompatibility test of Fbg-MP with C2C12 cell lines also revealed the close interactions, where the cells could grow around Fbg-MP without apoptosis. In conclusion, the EDC cross-linked Fbg-MPs were biodegradable and biocompatible with cells to be used as tissue scaffolds. Various chemokines,cytokines, drugs, and other peptides or proteins could also be incorporated in making Fbg-MP to be charged with diverse functionalities.
