| Abstract | Myricetin is a dietary flavonoid that is found in many fruits, vegetables, teas and medicinal herbs. It has been demonstrated to have antioxidative, antiapoptotic, and anti-inflammatory properties, but the precise mechanism underlying the effects of myricetin at the transcriptional level has not yet been determined. The present study aimed to determine the target genes of myricetin in human umbilical vein endothelial cells (HUVECs) by microarray gene expression profiling. Among the more than 40,000 genes investigated, 121 and 953 genes were upregulated by at least 1.5 fold, while 584 and 1426 genes were downregulated by at least 1.5 fold in 24 h for 25 關M and 100 關M myricetin treatment. Gene ontology (GO) analysis showed that the top biological processes disturbed in HUVECs were regulation of apoptosis and cellular metabolic processes. Furthermore, vascular diseaserelated genes including HIRA, HDAC9, HIF1A, and RTN3 were also altered by myricetin. These results demonstrate myricetin-induced genome wide alterations in the expression profile of HUVECs and suggest that myricetin exerts a vasculoprotective effect through transcriptional changes in the human endothelium. In conclusion, our data support the view that myricetinstimulated changes in gene expression contribute to protective effects on vascular disease. |
