Title Design of gold nanoparticle and DNA oligomer conjugates for enhancement or suppression of in vitro gene expression

Abstract Conjugates of gold nanoparticles and DNA oligomers (AuNP-DNA) were used in in vitro enhanced green fluorescent protein (eGFP) translation reaction. DNA sequences were designed to have varying binding energies to mRNA so that free DNA strands introduced to the reaction suppress gene expression in general. However, the amount of protein produced either increased or decreased when AuNP-DNA was used instead, depending on the concentration of AuNPDNA, coverage and the sequence of DNA strands. This result shows that AuNP-DNA?셲 specific or nonspecific adsorption to ribosomes, mRNA, and other bio-molecules can be tuned by an appropriate choice of DNA sequence, therefore AuNP-DNA can work as a biological entity that is either a gene silencing agent or a gene expression enhancer.
